All
the DNA molecules that are cut with a given
restriction enzyme will carry identical sequences
at their termini.

These
molecules, when mixed in the presence of the
enzyme DNA ligase, join with each other and
restore the restriction site.

The
rejoining can occur between the pieces that
were generated from the same DNA, or the
pieces that came from different DNA molecules.
The
latter are termed as ‘chimeric molecules’ or ‘recombinant
DNA’
